Diane Keaton a studiat drama la Colegiul Santa Ana College, inainte sa renunte, pentru a studia actoria la Scoala de Teatru Manhattan's Neighborhood Playhouse din New York.Dupa ce a devenit faimoasa cu cateva comedii ale lui Woody Allen, Diane Keaton merge mai departe incercand in lumea filmului atat ca actrita cat si ca regizor. Pe numele sau adevarat Diane Hall, vedeta a aparut in 1968 in piesa Hair.
In 1970, Diane Keaton debuteaza in film cu comedia Lovers and Other Strangers, dupa care apare alaturi de Al Pacino in 1972 in Nașul / The Godfather. In acelasi an ea si Woody Allen, de care Diane Keaton se indragosteste in viata reala, joaca in Play It Again, Sam si in 1973 joaca tot impreuna in Sleeper. Au urmat Nașul: Partea II / The Godfather Part II si Love and Death al lui Woody Allen.
Diane Keaton este una dintre cele mai bune actrite care reuseste si in cariera de regizor, astfel in 1995 debuteaza ca regizor cu drama Unstrung Heroes. A castigat un premiu al Academiei in 1977 pentru cea mai buna actrita in Anni Hall, si cu acelasi film si la aceeasi categorie a castigat un premiu al Academiei Britanice.
